This session will discuss a practical approach to bringing a new antibody into your laboratory. We will begin by determining how to best select a commercially available antibody. This is done by recognizing important details about that antibody prior to purchase, or in the case of “in house” developed antibodies understanding as much as possible about that antibody. All of the information gathered during this process will come in handy when initial protocol development is begun. In addition to selecting the appropriate antibody, extensive discussion on the selection of control tissues and control reagents will be reviewed. We will then learn how to determine the best initial approach that will include primary antibody concentrations, retrieval methods and what detection systems should be used. We will discuss how to evaluate the results and determine the appropriate “final” protocol and how to validate that protocol.
